Where do we come from? (What came before the Big Bang?)
Are we alone in the universe? OK you've kind of covered this, and not just in one universe either :-)
What happens when we die?
What is a good life?
How can we achieve world peace? Is it even possible?
Is there a God?
And more down to Earth, but still up in the sky :-) How come the moon fits exactly on top of the sun in an eclipse? Is that really just an enormous co-incidence?

Also, a kind of meta-question: Why should the origins of Life, the Universe, and Everything be such a mystery? Isn't that a mystery in itself?
